G1 Compartmentalization and Cell Fate Coordination
Pauklin and Vallier show that, whereas early G1 is permissive for TGF-β-dependent endoderm differentiation, cyclin D restricts the activity of Smad2/3 in late G1, resulting in a switch from endoderm to neuroectoderm potential in pluripotent stem cells. These findings provide insight into how signaling, the cell cycle, and lineage specification are coordinated.
متن کاملWhi5 phosphorylation embedded in the G1/S network dynamically controls critical cell size and cell fate
In budding yeast, overcoming of a critical size to enter S phase and the mitosis/mating switch--two central cell fate events--take place in the G1 phase of the cell cycle. Here we present a mathematical model of the basic molecular mechanism controlling the G1/S transition, whose major regulatory feature is multisite phosphorylation of nuclear Whi5. Cln3-Cdk1, whose nuclear amount is proportion...
متن کاملInvasive Cell Fate Requires G1 Cell-Cycle Arrest and Histone Deacetylase-Mediated Changes in Gene Expression.
Despite critical roles in development and cancer, the mechanisms that specify invasive cellular behavior are poorly understood. Through a screen of transcription factors in Caenorhabditis elegans, we identified G1 cell-cycle arrest as a precisely regulated requirement of the anchor cell (AC) invasion program. We show that the nuclear receptor nhr-67/tlx directs the AC into G1 arrest in part thr...
متن کاملPrespore cell fate bias in G1 phase of the cell cycle in Dictyostelium discoideum.
By generating a population of Dictyostelium cells that are in the G1 phase of the cell cycle we have examined the influence of cell cycle status on cell fate specification, cell type proportioning and its regulation, and terminal differentiation. The lack of observable mitosis during the development of these cells and the quantification of their cellular DNA content suggests that they remain in...
متن کاملCoordination of Cell Proliferation and Cell Fate Determination by CES-1 Snail
The coordination of cell proliferation and cell fate determination is critical during development but the mechanisms through which this is accomplished are unclear. We present evidence that the Snail-related transcription factor CES-1 of Caenorhabditis elegans coordinates these processes in a specific cell lineage. CES-1 can cause loss of cell polarity in the NSM neuroblast. By repressing the t...
